Histidyl-tRNA synthetase is a member of the aminoacyl-tRNA synthetase family, key enzymes of protein biosynthesis which charge tRNA molecules with the respective amino acids. This 58 kDa protein is an autoantigen recognized by Jo-1 antibodies which occur in 33% of patients with polymyositis and dermatomyositis. Jo-1 antibodies indicate an increased risk for lung involvement, and the disease tends to take a severe course with poor response to therapy.

Histidyl-tRNA synthetase (Jo-1) from DIARECT is expressed in the baculovirus expression system.

Additional antigenic targets of antibodies from the so-called anti-synthetase complex (PM/DM with autoantibodies against only a single aminoacyl-tRNA synthetase) are alanyl-tRNA synthetase (PL-12) and threonyl-tRNA synthetase (PL-7), respectively. In the subgroup of non-anti-Jo-1 cases PL-7 and PL-12 together have a prevalence of about 25%.
